Avame (ravimi)tööstuse mõlemad silmad
Kujutleme klaasi, mis on poolenisti täidetud vee ja poolenisti toiduõliga. Peal on õlikiht ja all on veekiht. Need kihid, või “faasid”, on üksteisest eraldatud. Samas, nad mõjutavad teineteist ning kõike, mis neis faasides toimub. Midagi taolist esineb ka inimrakkude välispiiril. Kui tarbime mõnda ravimit, näiteks ibuprofeeni, tavalist apteegi käsimüügis oleva valuvaigistit, siis selle osakesed jaotuvad meie analoogis veefaasi ja õlifaasi vahel ja on vastasmõjus mõlemaga. Paljuski sõltuvad need vastasmõjud keemiliste ühendite happelisusest – võimest loovutada vesinikioone ning samaaegselt omandada negatiivne laeng, teisiõnu, käituda happena. Selle protsessi käigus muutuvad ühendite omadused drastiliselt. Ibuprofeen on just üks selline ühend. Eksperimendid, mille põhjal ibuprofeeni ja teiste ühendite happelisust kirjeldatakse, viiakse enamasti läbi vaid ühes faasis. Kui tuleme tagasi meie klaasi juurde, siis see vastab justkui olukorrale, kus klaas on täidetud ainult veega. Samas on ibuprofeen lipofiilne ühend ning vee ja õli süsteemis liigub eeskätt just õlifaasi, kus ibuprofeeni omadused oluliselt erinevad omadustest veefaasis. Ühes faasis läbi viidud eksperiment ignoreerib seda olukorda täielikult. Seda võib võrrelda silmaklapi kandmisega – pool pilti läheb kaotsi. Miks selline pealiskaudne lähenemine niivõrd olulise rakenduse jaoks? Kuni viimase ajani ei olnud teoreetilist baasi ja eksperimentaalselt meetodit, mis võimaldaks mõõta ainetele usaldusväärseid happelisuse väärtusi tingimustes, mis on lähedasemad inimorganismis esinevale. Minu doktoritöö keskendus sellele, et selline baas ja meetod luua ning nüüd on inimkonnal olemas tööriistad selleks, et adekvaatsemalt hinnata ravimite, nagu ibuprofeen, ning paljude teiste keemiliste ühendite happelisusi ning sellest lähtuvalt ka muid omadusi erinevates olulistes rakendustes. Loodetavasti aitab see tõsta nii ravimitööstuse, või ka tervikuna võttes, tööstuse efektiivsust.
Expanding the vision of the (pharmaceutical) industry
Imagine a glass, half filled with cooking oil and half filled with water. On the top, there is the oil layer and, on the bottom, the aqueous layer. These layers, or phases, are visually separated from each other. But they still influence each other and the processes taking place in them. Something similar is present at the cell boundaries in every living organism. When a medicine, such as ibuprofen, a common over-the-counter painkiller, is consumed, it distributes between the water phase and the oil phase and interacts with both of them. Many of these interactions are influenced by the acidity of chemicals – the ability to donate a positively charged hydrogen atom to another molecule and become negatively charged, in other words, to act as an acid. As a result of this process, the properties of chemicals, such as ibuprofen, change dramatically. Experiments for measuring acidity of ibuprofen and other chemicals are almost always performed under monophasic conditions, something that in our glass example compares to a glass of just water. However, ibuprofen is a lipophilic compound, and in a water-oil system, it prefers the oil phase, where its properties are much different from its properties in an aqueous phase. Yet the interactions with the oil phase are completely ignored in a typical acidity measurement. It’s like seeing only half of the picture. Why is such an incomplete approach being used for something this important? Until recently, there was no theoretical basis or experimental method to conduct measurements of acidity under more relevant conditions. My doctor’s thesis focuses on providing the tools to measure acidity, and by extension, many other properties of molecules in biphasic systems in a more realistic and accurate way. Hopefully, these tools lead us to a more effective pharmaceutical industry, or industry as a whole.
