    Association of the COMT Val108/158Met genotype with professional career and education: The Val-allele is more frequent in managers and in enterprising occupations

    Catechol-O-methyl transferase (COMT) is a key player in neurotransmission by catecholamines, and the functional COMT Val108/158Met polymorphism is strongly related to prefrontal reactivity and to dopamine levels. As dopamine is a critically important neurotransmitter in cognition, emotion and motivation, we addressed the potential impact of this genotype on life course by examining its association with being in enterprising professions. The parents (n = 1410) of the target subjects in the Estonian Children Personality Behaviour and Health Study reported their current occupation, and those classified as enterprising (n = 197; 18%) were compared with the remaining group. Additionally, the subjects self-classified themselves according to the International Standard Classification of Occupations and the group of managers (6.2%) was compared to other groups. We found that the COMT Val108/158Met Val/Val homozygotes were overrepresented among enterprising occupations and the Val-allele carriers among self-classified managers. While several measures associated with the Val/Val homozygosity were also associated with enterprising occupation, no simple path from the genotype to enterprising occupations emerged from structural equation models, suggesting that the COMT Val108/158Met genotype contributes to choices of profession via multiple interactive features. We also reproduced a previous finding that the COMT genotype is associated with educational attainment in a gender-dependent manner.

    COMT Val158Met genotüübi seos söömishäirete sümptomite ja söömishäiretega

    Käesoleva uurimistöö eesmärk oli uurida Eesti laste isiksuse-, käitumise- ja terviseuuringu (ELIKTU) valimil, kas COMT Val158Met polümorfism on seotud söömishäirete sümptomite ning diagnoosiga. Töös leiti COMT peamõju söömishäire sümptomitele mõõdetuna EDI-2 küsimustikuga – Val/Val homosügootide skoorid kõhnuseihaluse ja kehaga rahulolematuse alaskaaladel olid kõrgemad kui Met/Met homosügootide omad. Lisaks leiti COMT x neurootilisus ja COMT x meelekindlus koosmõjud söömishäirete sümptomitele ning tuvastati soolised erinevused. Logistilise regressiooni tulemustest ilmnes, et Val/Val genotüüp ennustab söömishäire kujunemist. Uurimistöö panustab söömishäirete geneetilise aluse uurimisse ning annab suuna tulevasteks uuringuteks – hinnates COMT Val158Met polümorfismi mõju söömishäiretele, võiks arvesse võtta teisi katehhoolamiinide töötlust reguleerivaid geene ning negatiivseid elusündmusi.

    The influence of 5-HTTLPR, COMT Val158Met polymorphism, adverse life events, anxiety, impulsivity and neuroticism on eating disorder symptomatology

    The aim of this study was to examine the effect of 5-HTTLPR and COMT Val158Met polymorphism, anxiety, impulsivity, neuroticism and adverse life events on abnormal eating behaviors among 25-year-old women. This study is based on ECPBHS (Estonian Children Personality, Behaviour and Health Study) older cohort data. Participants answered to State and Trait Anxiety Inventory (STAI), Barrat Impulsiveness Scale (BIS-11), NEO-PI Estonian version, Eating Disorder Inventory -2 (EDI-2). The sample was genotyped for 5-HTTLPR and COMT Val158Met polymorphism. The main finding of the current study is that the influence of neuroticism on eating disorder symptomatology is mediated mainly by trait anxiety. This study shows consistent effects of neuroticism through trait anxiety on bulimic symptoms, body dissatisfaction and drive for thinness. Neuroticism through impulsivity influences only bulimic symptoms. Trait anxiety can be seen as a stable trait predisposing people toward higher levels of eating disorder symptomatology.
